Extending the Applicability of Using Ternary Reversed-Phase Systems

Recent work in our laboratory demonstrated that ternary reversed-phase solvent systems (water-acetonitrile-methanol) could be optimized with a small number of experiments. This presentation will show additional applications for this technique that further demonstrate the utility and convenience of using ternary solving combinations. More demanding applications such as cannabis compounds and pharmaceutical impurities demonstrate that a ternary solvent system provides an easy procedure for finding the optimum conditions. The technique also finds additional uses in related fields such as method validation.
One of the many challenges of method development is demonstrating that the final method does not suffer from coeluting impurities. Tests for “Specificity” or the search for an “Orthogonal Method” are important experiments that can be time-consuming and expensive. For many separations, ternary mobile phases can be used to clearly demonstrate specificity or orthogonality. With only three experiments it is possible to track both main ingredients and impurities to confirm that certain interferences are not occurring. We will show real-world examples for each of these ideas.

Dr. Merlin K. L. Bicking (Presenter) is President and Senior Analytical Scientist, ACCTA, Inc. He has extensive analytical chemistry experience in academia, contract research, independent testing laboratories, consulting, and technical training. His professional history includes the development of two EPA methods, as well as numerous methods in other regulated and non-regulated industries. His publications and presentations cover a wide range of topics, including liquid chromatography theory, derivatization, method optimization, and the use of experimental design strategies in analytical chemistry. He also develops and presents technical training seminars for analytical laboratory staff.
